Right, I dissolve 39g potato-dextrose with 5g agar in one liter water, I adjust the ph, I sterilize it for 20 min, I pour it in 20 dishes, I wait a couple of days to see if I worked sterile enough, then I innoculate them :ph34r: If you do not wait, you have chance to innoculate a petri which isn't sterile anymore...

The biologist I work with told me to do so, he works with tissue-culture (orchids, meat-eaters, aquarium plants and much more)

I use ready made potato dextrose agar, I sterilize it for 20 minutes then I leave the fresh dishes in the incubator for 4-5 days to see if they are poured clean.

How do you obtain a clean culture? I tried to select apparently clean mycelium to inoculate a new petri dish, but I always carry some pennicillium or something like that; should I bath the wedge in a peroxide solution? I thought It shouldn't be neccessary   :huh:

I went out to water the azurescens bed and I found a cluster of 5 mushrooms pinning ^_^
So I gentley watered around the cluster :ph34r:
Will there come more as first flush? the bed is 1 square meter :huh:

Does anyone here make tissue-culture?
I did my first try a couple of days ago.
I used an opuntia to learn the process, if project goes fine I'll make a pile of trichocereus :ph34r:

The surface of the bed is hardly colonized but it shows a blood colour secretion, is that mycelium piss? mycelium is direct in contact with a plastic bag, I thought it was missing fresh air, so I've put a page drawning paper between mycelium and plastic hoping is breathes better  :huh:

Beds are 100% done, should I enlarge them?
It'sT now 15kg woodchips, 1:5 becomes 75kg fresht chipst :ph34r:

Maybe the myc has to establish to make a unique network  so I should leave the bedsT as they are :huh:

Use your pc, microwave does not reach the right temperature to sterilize grains, I use the microwave to sterilize the casing mix, misting and syringe water, agar and  not much more.

If you have contam in your jars, you must increase sterilization time.
