Cultivation / Re: Agar cleaning process

Right, I dissolve 39g potato-dextrose with 5g agar in one liter water, I adjust the ph, I sterilize it for 20 min, I pour it in 20 dishes, I wait a couple of days to see if I worked sterile enough, then I innoculate them  If you do not wait, you have chance to innoculate a petri which isn't sterile anymore...

The biologist I work with told me to do so, he works with tissue-culture (orchids, meat-eaters, aquarium plants and much more)

I use ready made potato dextrose agar, I sterilize it for 20 minutes then I leave the fresh dishes in the incubator for 4-5 days to see if they are poured clean.

How do you obtain a clean culture? I tried to select apparently clean mycelium to inoculate a new petri dish, but I always carry some pennicillium or something like that; should I bath the wedge in a peroxide solution? I thought It shouldn't be neccessary

So I gentley watered around the cluster
Will there come more as first flush? the bed is 1 square meter

I'll use them to sterilize grains, colonize pasteurized dung-straw substrates and as fruiting chamber for cube

Does anyone here make tissue-culture?
I did my first try a couple of days ago.
I used an opuntia to learn the process, if project goes fine I'll make a pile of trichocereus

Mycelium looks thin as cobweb but rhyzomorph, think it's doing fine, am I right?

The surface of the bed is hardly colonized but it shows a blood colour secretion, is that mycelium piss? mycelium is direct in contact with a plastic bag, I thought it was missing fresh air, so I've put a page drawning paper between mycelium and plastic hoping is breathes better

Got the plug, thanks!
shall I inoculate fine woodchips or cardboard?

Beds are 100% done, should I enlarge them?
It'sT now 15kg woodchips, 1:5 becomes 75kg fresht chipst

Maybe the myc has to establish to make a unique network  so I should leave the bedsT as they are

Hi! After 1 week my two jar of birth one contaminated with green ai ai...for first attempt i think its ok.....the luky jar has a beginning of micelium....after 6 days....i think the spore syringe was not full o spores....boh

but i have a question for the Masters.....i have a little pressure can i do to preparate many jars? or better....can i sterilize my jars with microwave? it works?
Can i coock my grain apart...and then fill the jar and then sterilize in microwave?

Thanx to all......and sorry for my language...

Use your pc, microwave does not reach the right temperature to sterilize grains, I use the microwave to sterilize the casing mix, misting and syringe water, agar and  not much more.

If you have contam in your jars, you must increase sterilization time.

you cannot be an anarchist in this job -_-

Very nice, how do you make them? Do you have a program to develop them?
