Can squirting spores on pasturized straw and horse shit work directly?

Spores germinate very slowy directly on bulk substrate.
It is not impossible, but it is much better if you use spawn.

So then it is possible, so can use a plastic zip-lock filled with pasturized straw,horse substrate and inject it with spores and work? And how long does it usually take in order for the substrate to fully colonize?

Try it and report.
It is difficult to estimate the colonization times, but as I said, it could take a very long time, several weeks perhaps. I deffinitley suggest to get some spawn going first and inocukate with it.
