I did 3 jars originally and I felt 2 were suspect. I went with that jar as the mother and got good growth. This was the spore inoc jar rather than the bfr myc jars.

I was absolutely positive it was mycelium and not contam after many hours of picture research.

I then made two more 'user jars' from the mother and inocced half my jars from the best one of these.

The other half was from the original mother jar as it was in the syringe already.

I've had zero growth out of all jars and no contams seen either. It's no been 15 days, something has clearly gone wrong.  :huh:

Any ideas?

My first LC went great but I would say do a few jars at the beginning so you don't have all your eggs in 1 basket.

I also homebrew so the dextros/malt beer enhanser was my medium of choice. I used my hydrometer for an accurate 4% sugar solution (SG 1.015)

You'll see growth in 2-7 days and be ready to inoc jars in 6-10.

I inoculated with LC a week ago. The LC was thick and good but I still haven't spotted any growth yet.

Temps have been kept at 23-27C so I would of thought with liquid culture it should of shown signs in a couple of days.

Thanks for your reply dub,

I've refrigerated them thinking if any contams did crop up, the sterilisation would kill it before the inoc.
In retrospect the sterilise now method would knock out potential baddies right at the start.

I may have too go with what I've done and see what pans out.

I'm inoculating with LC so as soon as its ready I'm there. The way its progressing there may be more than enough to use before 6 days.

Large capacity needles would be a def bonus though. I want to inoc jars with 4-5cc each (1-1.25cc each site)  so will have to return for a refill 3 times with my 10ml needle, to my LC user jar, potentially increasing the chance of contams in both LC jar and brf cakes.

Just transferred 10ml of multispore 'mother' LC multispore solution split into 2 user LC jars.

The transfer went beautifully the 2 x daily agitation with glass shards worked a treat and the myc didn't block the syringe at all.

This should mean I only ever have to go back into my 'mother' if both my user LC's contaminate.

I've refrigerated the mother to keep her good. She should last 6-12 months.

Hope fully one of them should be contam free at least and be ready to inoc my second batch of BRF cakes.

I used the standard 4% in solution but less malt in the mix. 1% malt 3% dextros this time to make clarity and observation a little easier.

For any one interested in critical measuring not wanting to use scales you can you a brewing hydrometer. Only a couple of quid (UK)
Specific Gravityon or SG on a brewing hydrometer of a 4% sugar solution is 1.015. Plain water should read 1.000 so adjustments must be made for water inaccuracy.
This is what I've worked out as I don't have scales and it's worked great.

With the amount of myc in each of the new medium and according to my projected growth rate based on the mother LC. I think they'll be ready to use in 6 days.

I've made a new batch of BRF cakes but realised I won't be ready to inoc for another 6-9 days.

Do I refrigerate now then sterilise?

or

Sterilise now then refrigerate and bring to room temp before inoc?

Could be Feb - Sep depending on your climate.

I'm UK so not familiar with Mid West climate.

It fruits near 100% humidity and temperature ranges from 80-86 degrees Fahrenheit. So let it rain for a day or two when its warm then go hunting.

My multispore inoculate LC jar is wicked.

Its 13 days and fully thick with myc in clear malt dextros medium. Incubator temps have been a good 76f

The other 2, started well but I think they were over run. Cloudy and sediment difficult to see what is going on.

I'm going to take this 'mother' and make a second LC for more frequent use from it.

I'm going to use 1% malt 3% dextros this time to make clearity and observation easier.

I'll then refrigerate it for eternity.  :wacko:

I figured it was an account issue why I couldn't view pics but if you go through google images you can see them.  :ph34r:

2 are cloudy now and looks like they have been taken over since I can't see any frog spawn like clumps.
The last looks great. With 50%+ clumps by volume in suspension. The malt-dextros is crystal clear yellow. It looks beautiful.

I think this will become the mother although it's mixed spore rather than clone or isolate.
I'll try to get a pic up.

2 jars inoculated with 1cc of spores each. One looks cloudy with stringy white bits that float. The other has 50% frog spawn like floating in beautiful clumps in clear malt fluid.
1 jar inoculated with fluid injected then withdrawn from full colonized PF cake also gone cloudy with bits.

I'm sure the cloudy ones are contams but the clear one with frog like spawn is awsome.

Will growth continue much more?
This is day 10 of the LC tek and my temps are 76-80f.

Any info wold be great.

It took my spores 11 days to germinate and only 1 out of 24 inoc sites across 6 jars came good.

I cooked them for 2 hours @ 35C (95F) by mistake right at the very beginning after inoculation.

Hang in there. Re inoculation may be risky if your cakes are drying out already as they won't reach full colonization.

If you do choose to re inoculate I would do it with fresh prepared substrate so you know the water ratio is good.

yeah ...the whole cake man.

A fully colonized cake is a strong one resistant to bacterial infection once you birth it to your fruiting chamber.

If you used bigger jars they can some times dry out and not colonize. Keep an eye on your temps so you don't over heat and therefore dry out your cakes.

The other thing about bigger jars for cakes is that the longer colonization can mean a denser build up of CO2 at the bottom of your jar, which can inhibit myc growth.
As growth slows down you can try inverting the jars to dissipate the CO2 allowing the myc to colonize the last bit at the bottom.

Its not a race done proper is better than faster.

Both work but you can see though glass better so easily spot any contams early. If you do use plastic make sure it can take the heat from sterilisation and the extra hot heat from pressure cookers in the future may be.

-i most likely wont be buying a pressure cooker, ill probably just boil/steam the jars.  another thing im not 100% on.  when putting them in the pot should i leave just the top on the container with the holes, or should there be something over that?  A layer of foil over so steam will not penatrate and ruin yor water ratios.  id assume something over it to make sure no contaminants get in, but just want to make sure.  also if i decide to go with the plastic how would that work with the steaming?  it wont warp the container as long as its strong enough? Must be high temp thermo plastic

-while using the 1/2 pint containers what is the best mix of substrate?  ive seen a lot of 1/2 cup verm &1/6 cup BRP, but it was unclear how much water to use for that mix.  also 1/8 cup BRP, 1/2-2/3 cup verm, and 3 tablespoons water, is that presice? this claims to be the maximum fruting formula: 1/4 cup BRP, 1/2 cup verm, and 60 cc's water. any suggestions of the "perfect" mixture for the 1/2 container? 

1/2 pint BRF cakes 2:1:1 Verm:BRF:H20. slightly too dry is better than too wet. Loose packed and dry verm top layer

-ive read a lot that the dunk helps them grow bigger and gives more flushes.  im not sure i undertand the flushing 100%.  wouldnt that give more chance of contamination with all the dunking and touching and rolling and what not?  after the dunking is it as usual then with the putting it on the perlite and everything after?

Once a cake is 100% fully colonized its strong and can easily keep bad contams from it food source. its a long time to be in the jar so the dunk helps hydrate the cake and refil the tanks for the next stage. Shrooms are 90% water and that has to come from some where. After dunkin' put your babies on a lid or foil on the perlite. The dunk is for the rehydration of the cake, the perlite to keep humidity up.

-after the dunk or removal from container im not sure how close or far apart the the cakes should be in the tub ill be putting the in.  i saw a lot of keep them close-ish as to have the humidity from eachother help keep humid, i also heard far apart to make sure they dont fuck with eachother.  input?

Enough room to let 'shrooms grow out all ways but not touch each other. 3 inches should be OK

-lighting.   ive read a lot about using flourescent lights, but then again i see a lot that they do not need too much direct light...maybe an hour or so a day of natural sunlight, otherwise keep pretty dark.  what is and is not true for them?  do they need a lot of lighting from a flourescent, or can i keep them in a closet and light just a little bit a day?  or can i just keep the cakes in  a in a clear-ish plastic tub with a clearish top?  also ive read that keeping the inside of the terranium dark so they grow strait up is better, but would keeping it clear all around yield a bigger growth bc the light would be putting them in all directions or no?

Natural cycle light is fine. If your project is stealth a few hours a day is fine. Making the sides only dark of the vivarium will make it automaticly lighter at the top. If FAE is good and your humidity is good then they will be straighter. I like the wilder groove since we aint growing soldiers but it can make harvest better. -_-

-i know they need fresh air exchange and humidity.  this kind of goes in hand with the lighting question. if i am keeping them in a closet and bringing them out every few hours to spray, is the fanning and fresh air exchange like that good enough or no?

2x daily, more if you can

sorry theres so much, i just want to make sure i do this as correctly as possible.

Start simple for you first grow then think about lights etc once you get the bug and wanna tweak your methods.

If your room temp is healthy 70f I wouldn't bother heating. but insulation will help to retain there own heat. Those babies will get there in the end it isn't a race.

Contams flourish in warmer heat more than myc growth so if your cleanliness ain't up to scratch you might just encourage bad stuff to grow and take over your project before you white stuff has even got a foot hold.

Your mycelium is a simple living thing. Constant is much better than warm cold warm fluctuation.

Many people just pop jars into a cooler box unheated to reduce fluctuation bt even bubble wrap would be better than none.

It seems to have all worked.

Day 53 since inoculation.

42 days of myc growth after 11 day germination and a dry stall.

10-05-11:  Injected 30cc malt dextrose 4% solution – extracted 5cc for LC
12-05-11:  Inverted jar to drain any excess moisture and help CO2 sink away.
13-05-11:  Colonizing bottom now 95% fully colonized
16-05-11:  Colonized now 99% fully. Birth to dunk 'n'roll 17th

I'm gonna birth and hydrate my cake for 24hrs then roll in dry verm and place in a gallon pod bottle chamber.

Since I got only one cake this time round I'm gonna give it the personal attention it deserves and my LC's are all coming on great.

I fucked up big time at the beginning :huh: :rolleyes: but I kept my cool  :mellow: and made a plan and my patience.

53 days for a cake aint the quickest but this one will be remembered as the Get Out of Jail Tek  ^_^

I'm growing 3 liquid cultures.

Extra Light Malt and Dextros
6 tsp sugars in 750ml solution (4% = 30g = 720ml)
2g sugar per 1 point mark on brewing hydrometer in a 750ml bottle
4% solution = 15 points SG:1.015

3 Jars half full of medium + glass shards.
Steam boiled for 30 mins
2 jars inoculated with 1cc of spores each
1 jar inoculated with fluid injected then withdrawn from full colonized PF cake

2 small piece of broken glass were added to the jar before sterilization. Agitating daily to cut up the mycellium which can form into an unsuckable clump in the jar

These are my questions:

The medium is a light brown. Will the growth still be white?
As the myc grows does it naturally fall to the bottom of the jar in between agitation or should it be more floaty?
